Plays a key role in the instruction of early lymphoidprogenitors to develop into B lineage by repressing T-cellinstructive Notch signals (By similarity). Specifically repressesthe transcription of the CDKN2A gene. Efficiently abrogates E2F1-dependent CDKN2A transactivation/de-repression. Binds to theconsensus sequence 5'-[GA][CA]GACCCCCCCCC-3' (By similarity).{ECO:0000250}. Antagonizes canonical Wnt signaling by inhibiting LRP5/6interaction with Wnt and by forming a ternary complex with thetransmembrane protein KREMEN that promotes internalization ofLRP5/6. DKKs play an important role in vertebrate development,where they locally inhibit Wnt regulated processes such as antero-posterior axial patterning, limb development, somitogenesis andeye formation. In the adult, Dkks are implicated in bone formationand bone disease, cancer and Alzheimer disease (By similarity).{ECO:0000250}.
