Acyl-CoA thioesterases are a group of enzymes thatcatalyze the hydrolysis of acyl-CoAs to the free fatty acid andcoenzyme A (CoASH), providing the potential to regulateintracellular levels of acyl-CoAs, free fatty acids and CoASH. Hasacyl-CoA thioesterase activity towards medium (C12) and long-chain(C18) fatty acyl-CoA substrates. Can also hydrolyze 3-hydroxyphenylacetyl-CoA and 3,4-dihydroxyphenylacetyl-CoA (invitro). May play a role in controlling adaptive thermogenesis (Bysimilarity). {ECO:0000250, ECO:0000269|PubMed:16934754,ECO:0000269|PubMed:19170545}. Plays a role in influencing the retrograde traffickingof lysosomal sorting receptors SORT1 and IGF2R from the endosomesto the trans-Golgi network by controlling the recruitment ofretromer complex to the endosomal membrane. Regulates thelocalization and activation of RAB7A which is required to recruitthe retromer complex to the endosomal membrane (PubMed:22431521).{ECO:0000269|PubMed:22431521}.
