Serine/threonine-protein kinase involved in autophagy inresponse to starvation. Acts upstream of phosphatidylinositol 3-kinase PIK3C3 to regulate the formation of autophagophores, theprecursors of autophagosomes. Part of regulatory feedback loops inautophagy: acts both as a downstream effector and negativeregulator of mammalian target of rapamycin complex 1 (mTORC1) viainteraction with RPTOR. Activated via phosphorylation by AMPK andalso acts as a regulator of AMPK by mediating phosphorylation ofAMPK subunits PRKAA1, PRKAB2 and PRKAG1, leading to negativelyregulate AMPK activity. May phosphorylate ATG13/KIAA0652 andRPTOR; however such data need additional evidences. Plays a roleearly in neuronal differentiation and is required for granule cellaxon formation. May also phosphorylate SESN2 and SQSTM1 toregulate autophagy (PubMed:25040165).{ECO:0000269|PubMed:18936157, ECO:0000269|PubMed:21460634,ECO:0000269|PubMed:21795849, ECO:0000269|PubMed:25040165}. Substrate-recognition component of a Cul7-RINGubiquitin-protein ligase complex, which mediates theubiquitination and subsequent proteasomal degradation of targetproteins. The Cul7-RING(FBXW8) complex mediates ubiquitination andconsequent degradation of GORASP1, acting as a component of theubiquitin ligase pathway that regulates Golgi morphogenesis anddendrite patterning in brain (PubMed:21572988). Mediatesubiquitination and degradation of IRS1 in a mTOR-dependent manner:the Cul7-RING(FBXW8) complex recognizes and binds IRS1 previouslyphosphorylated by S6 kinase (RPS6KB1 or RPS6KB2)(PubMed:18498745). The Cul7-RING(FBXW8) complex also mediatesubiquitination of MAP4K1/HPK1: recognizes and bindsautophosphorylated MAP4K1/HPK1, leading to its degradation,thereby affecting cell proliferation and differentiation(PubMed:24362026).
