Receptor tyrosine kinase that transduces signals fromthe extracellular matrix into the cytoplasm by binding to severalligands including TULP1 or GAS6. Regulates many physiologicalprocesses including cell survival, migration and differentiation.Ligand binding at the cell surface induces dimerization andautophosphorylation of TYRO3 on its intracellular domain thatprovides docking sites for downstream signaling molecules.Following activation by ligand, interacts with PIK3R1 and therebyenhances PI3-kinase activity. Activates the AKT survival pathway,including nuclear translocation of NF-kappa-B and up-regulation oftranscription of NF-kappa-B-regulated genes. TYRO3 signaling playsa role in various processes such as neuron protection fromexcitotoxic injury, platelet aggregation and cytoskeletonreorganization. Plays also an important role in inhibition ofToll-like receptors (TLRs)-mediated innate immune response byactivating STAT1, which selectively induces production ofsuppressors of cytokine signaling SOCS1 and SOCS3.{ECO:0000269|PubMed:20546121}. (Microbial infection) Acts as a receptor for lassa virusand lymphocytic choriomeningitis virus, possibly through GAS6binding to phosphatidyl-serine at the surface of virion envelope.{ECO:0000269|PubMed:22156524, ECO:0000269|PubMed:22673088,ECO:0000269|PubMed:25277499}. (Microbial infection) Acts as a receptor for Ebolavirus,possibly through GAS6 binding to phosphatidyl-serine at thesurface of virion envelope. {ECO:0000269|PubMed:17005688}. | TJP1, TJP2, and TJP3 are closely related scaffoldingproteins that link tight junction (TJ) transmembrane proteins suchas claudins, junctional adhesion molecules, and occludin to theactin cytoskeleton (PubMed:7798316, PubMed:9792688). The tightjunction acts to limit movement of substances through theparacellular space and as a boundary between the compositionallydistinct apical and basolateral plasma membrane domains ofepithelial and endothelial cells. Necessary for lumenogenesis, andparticularly efficient epithelial polarization and barrierformation (By similarity). Plays a role in the regulation of cellmigration by targeting CDC42BPB to the leading edge of migratingcells (PubMed:21240187). Plays an important role in podosomeformation and associated function, thus regulating cell adhesionand matrix remodeling (PubMed:20930113).
