Part of the CASK/TBR1/TSPYL2 transcriptional complexwhich modulates gene expression in response to neuronal synapticactivity, probably by facilitating nucleosome assembly. Mayinhibit cell proliferation by inducing p53-dependent CDKN1Aexpression. {ECO:0000269|PubMed:11395479,ECO:0000269|PubMed:17317670}. Deubiquitinating enzyme involved in beta-2 adrenergicreceptor (ADRB2) recycling. Acts as a regulator of G-proteincoupled receptor (GPCR) signaling by mediating thedeubiquitination beta-2 adrenergic receptor (ADRB2). Plays acentral role in ADRB2 recycling and resensitization afterprolonged agonist stimulation by constitutively binding ADRB2,mediating deubiquitination of ADRB2 and inhibiting lysosomaltrafficking of ADRB2. Upon dissociation, it is probablytransferred to the translocated beta-arrestins, possibly leadingto beta-arrestins deubiquitination and disengagement from ADRB2.This suggests the existence of a dynamic exchange between theADRB2 and beta-arrestins. Deubiquitinates DIO2, thereby regulatingthyroid hormone regulation. Deubiquitinates HIF1A, leading tostabilize HIF1A and enhance HIF1A-mediated activity. Mediatesdeubiquitination of both 'Lys-48'- and 'Lys-63'-linkedpolyubiquitin chains. {ECO:0000269|PubMed:12056827,ECO:0000269|PubMed:12865408, ECO:0000269|PubMed:15776016,ECO:0000269|PubMed:19424180}.
