TOP3A

Q13472

May play a role in the regulation of keratinocytedifferentiation. Releases the supercoiling and torsional tension of DNAintroduced during the DNA replication and transcription bytransiently cleaving and rejoining one strand of the DNA duplex.Introduces a single-strand break via transesterification at atarget site in duplex DNA. The scissile phosphodiester is attackedby the catalytic tyrosine of the enzyme, resulting in theformation of a DNA-(5'-phosphotyrosyl)-enzyme intermediate and theexpulsion of a 3'-OH DNA strand. The free DNA strand thenundergoes passage around the unbroken strand thus removing DNAsupercoils. Finally, in the religation step, the DNA 3'-OH attacksthe covalent intermediate to expel the active-site tyrosine andrestore the DNA phosphodiester backbone. Essential component ofthe RMI complex, a complex that plays an important role in theprocessing of homologous recombination intermediates to limit DNAcrossover formation in cells. Has DNA decatenation activity.{ECO:0000269|PubMed:20445207, ECO:0000269|PubMed:8622991}.
