Acts as a regulator of the tubular endoplasmic reticulum(ER) network. Suppresses the RTN3/4-induced formation of the ERtubules (PubMed:25612671). Positively regulates PERK-mediated andIRE1-mediated unfolded protein response signaling(PubMed:26268696). {ECO:0000269|PubMed:25612671,ECO:0000269|PubMed:26268696}. E3 SUMO-protein ligase; has a preference for SUMO2 andSUMO3 and facilitates UBE2I/UBC9-mediated sumoylation of targetproteins (PubMed:26524493, PubMed:26524494). Plays a role inprotein SUMO2 modification in response to stress caused by DNAdamage and by proteasome inhibitors (in vitro). Required for MCM4sumoylation (By similarity). Has no activity with SUMO1(PubMed:26524493). Preferentially transfers an additional SUMO2chain onto the SUMO2 consensus site 'Lys-11' (PubMed:26524493).Negatively regulates transcriptional activation mediated by theSMAD4 complex in response to TGF-beta signaling.
