Modulates the activity of RHO-like proteins and connectsextracellular signals to cytoskeletal activities. Acts as a GDP-dissociation stimulator protein that stimulates the GDP-GTPexchange activity of RHO-like GTPases and activates them. Mediatesextracellular laminin signals to activate Rac1, contributing toneurite growth. Involved in lamellipodial formation andadvancement of the growth cone of embryonic hippocampal neurons.Promotes migration of neurons in the cerebral cortex. Whenoverexpressed, induces membrane ruffling accompanied by theaccumulation of actin filaments along the altered plasma membrane(By similarity). Activates specifically RAC1, but not CDC42 andRHOA. {ECO:0000250, ECO:0000269|PubMed:10512681}. Component of the 9-1-1 cell-cycle checkpoint responsecomplex that plays a major role in DNA repair (PubMed:10846170,PubMed:10884395). The 9-1-1 complex is recruited to DNA lesionupon damage by the RAD17-replication factor C (RFC) clamp loadercomplex (PubMed:12578958). Acts then as a sliding clamp platformon DNA for several proteins involved in long-patch base excisionrepair (LP-BER) (PubMed:15871698). The 9-1-1 complex stimulatesDNA polymerase beta (POLB) activity by increasing its affinity forthe 3'-OH end of the primer-template and stabilizes POLB to thosesites where LP-BER proceeds; endonuclease FEN1 cleavage activityon substrates with double, nick, or gap flaps of distinctsequences and lengths; and DNA ligase I (LIG1) on long-patch baseexcision repair substrates (PubMed:15314187, PubMed:15556996,PubMed:15871698). The 9-1-1 complex is necessary for therecruitment of RHNO1 to sites of double-stranded breaks (DSB)occurring during the S phase (PubMed:21659603).
