Acts as a transcriptional repressor. Interacts with MXI1to repress MYC responsive genes and antagonize MYC oncogenicactivities. Interacts with MAD-MAX heterodimers by binding to MAD.The heterodimer then represses transcription by tethering SIN3B toDNA. Also forms a complex with FOXK1 which repressestranscription. With FOXK1, regulates cell cycle progressionprobably by repressing cell cycle inhibitor genes expression.{ECO:0000250|UniProtKB:Q62141}. Probable GTPase-activating protein for Rab8; itstransient association with Rab8 is mediated by OPTN. InhibitsRab8-mediated endocytic trafficking, such as of transferrinreceptor (TfR) and reduces Rab8 recruitnment to tubules emanatingfrom the endocytic recycling compartment (ERC). Involved inregulation of autophagy. Mediates inhibition of autophagy causedby the OPTN variant GLC1E LYS-50; the function requires itscatalytic activity, however, the involved Rab is not known.{ECO:0000269|PubMed:22854040, ECO:0000269|PubMed:24752605}.
