TXK

P42681

PKC-related serine/threonine-protein kinase and Rho/Raceffector protein that participates in specific signal transductionresponses in the cell. Plays a role in the regulation of cellcycle progression, actin cytoskeleton assembly, cell migration, cell adhesion, tumor cell invasion and transcription activationsignaling processes. Phosphorylates CTTN in hyaluronan-inducedastrocytes and hence decreases CTTN ability to associate withfilamentous actin. Phosphorylates HDAC5, therefore lead to impairHDAC5 import. Direct RhoA target required for the regulation ofthe maturation of primordial junctions into apical junctionformation in bronchial epithelial cells. Required for G2/M phasesof the cell cycle progression and abscission during cytokinesis ina ECT2-dependent manner. Stimulates FYN kinase activity that isrequired for establishment of skin cell-cell adhesion duringkeratinocytes differentiation. Regulates epithelial bladder cellsspeed and direction of movement during cell migration and tumorcell invasion. Inhibits Akt pro-survival-induced kinase activity.Mediates Rho protein-induced transcriptional activation via the c-fos serum response factor (SRF). Phosphorylates HCV NS5B leadingto stimulation of HCV RNA replication. Involved in the negativeregulation of ciliogenesis (PubMed:27104747).{ECO:0000269|PubMed:10226025, ECO:0000269|PubMed:10926925,ECO:0000269|PubMed:11777936, ECO:0000269|PubMed:11781095,ECO:0000269|PubMed:15364941, ECO:0000269|PubMed:17332740,ECO:0000269|PubMed:20188095, ECO:0000269|PubMed:20974804,ECO:0000269|PubMed:21754995, ECO:0000269|PubMed:27104747,ECO:0000269|PubMed:9121475}. Non-receptor tyrosine kinase that plays a redundant rolewith ITK in regulation of the adaptive immune response. Regulatesthe development, function and differentiation of conventional T-cells and nonconventional NKT-cells. When antigen presenting cells(APC) activate T-cell receptor (TCR), a series of phosphorylationlead to the recruitment of TXK to the cell membrane, where it isphosphorylated at Tyr-420. Phosphorylation leads to TXK fullactivation. Contributes also to signaling from many receptors andparticipates in multiple downstream pathways, including regulationof the actin cytoskeleton. Like ITK, can phosphorylate PLCG1,leading to its localization in lipid rafts and activation, followed by subsequent cleavage of its substrates. In turn, theendoplasmic reticulum releases calcium in the cytoplasm and thenuclear activator of activated T-cells (NFAT) translocates intothe nucleus to perform its transcriptional duty. With PARP1 andEEF1A1, TXK forms a complex that acts as a T-helper 1 (Th1) cell-specific transcription factor and binds the promoter of IFNG todirectly regulate its transcription, and is thus involvedimportantly in Th1 cytokine production. Phosphorylates both PARP1and EEF1A1. Phosphorylates also key sites in LCP2 leading to theup-regulation of Th1 preferred cytokine IL-2. Phosphorylates 'Tyr-201' of CTLA4 which leads to the association of PI-3 kinase withthe CTLA4 receptor. {ECO:0000269|PubMed:10523612,ECO:0000269|PubMed:11564877, ECO:0000269|PubMed:11859127,ECO:0000269|PubMed:17177976, ECO:0000269|PubMed:9813138}.
