Serine/threonine kinase which acts as a negativeregulator of Ras-related Rap2-mediated signal transduction tocontrol neuronal structure and AMPA receptor trafficking. Requiredfor normal synaptic density, dendrite complexity, as well assurface AMPA receptor expression in hippocampal neurons. Canactivate the JNK and MAPK14/p38 pathways and mediates stimulationof the stress-activated protein kinase MAPK14/p38 MAPK downstreamof the Raf/ERK pathway. Phosphorylates: TANC1 upon stimulation byRAP2A, MBP and SMAD1. Has an essential function in negativeselection of thymocytes, perhaps by coupling NCK1 to activation ofJNK1. Isoform 4 can activate the JNK pathway. Involved in theregulation of actin cytoskeleton reorganization, cell-matrixadhesion, cell-cell adhesion and cell migration. Non-heme iron-containing dioxygenase that catalyzes thestereo-specific peroxidation of free and esterifiedpolyunsaturated fatty acids generating a spectrum of bioactivelipid mediators. Mainly converts arachidonic acid to (12S)-hydroperoxyeicosatetraenoic acid/(12S)-HPETE but can alsometabolize linoleic acid. Has a dual activity since it alsoconverts leukotriene A4/LTA4 into both the bioactive lipoxinA4/LXA4 and lipoxin B4/LXB4. Through the production of specificbioactive lipids like (12S)-HPETE it regulates differentbiological processes including platelet activation. It alsoprobably positively regulates angiogenesis through regulation ofthe expression of the vascular endothelial growth factor. Plays arole in apoptotic process, promoting the survival of vascularsmooth muscle cells for instance.
