Binds peptides derived from antigens that access theendocytic route of antigen presenting cells (APC) and presentsthem on the cell surface for recognition by the CD4 T-cells. Thepeptide binding cleft accommodates peptides of 10-30 residues. Thepeptides presented by MHC class II molecules are generated mostlyby degradation of proteins that access the endocytic route, wherethey are processed by lysosomal proteases and other hydrolases.Exogenous antigens that have been endocytosed by the APC are thusreadily available for presentation via MHC II molecules, and forthis reason this antigen presentation pathway is usually referredto as exogenous. As membrane proteins on their way to degradationin lysosomes as part of their normal turn-over are also containedin the endosomal/lysosomal compartments, exogenous antigens mustcompete with those derived from endogenous components. Autophagyis also a source of endogenous peptides, autophagosomesconstitutively fuse with MHC class II loading compartments. Inaddition to APCs, other cells of the gastrointestinal tract, suchas epithelial cells, express MHC class II molecules and CD74 andact as APCs, which is an unusual trait of the GI tract. To producea MHC class II molecule that presents an antigen, three MHC classII molecules (heterodimers of an alpha and a beta chain) associatewith a CD74 trimer in the ER to form a heterononamer. Soon afterthe entry of this complex into the endosomal/lysosomal systemwhere antigen processing occurs, CD74 undergoes a sequentialdegradation by various proteases, including CTSS and CTSL, leavinga small fragment termed CLIP (class-II-associated invariant chainpeptide). The removal of CLIP is facilitated by HLA-DM via directbinding to the alpha-beta-CLIP complex so that CLIP is released.HLA-DM stabilizes MHC class II molecules until primary highaffinity antigenic peptides are bound. The MHC II molecule boundto a peptide is then transported to the cell membrane surface. InB-cells, the interaction between HLA-DM and MHC class II moleculesis regulated by HLA-DO. Primary dendritic cells (DCs) also toexpress HLA-DO. Lysosomal microenvironment has been implicated inthe regulation of antigen loading into MHC II molecules, increasedacidification produces increased proteolysis and efficient peptideloading. Involved in pre-mRNA splicing as component of thespliceosome (PubMed:11991638, PubMed:28502770, PubMed:28076346).Is required in the specific splicing of CDKN1A pre-mRNA; thefunction probably involves the recruitment of U2AF2 to the mRNA.Is proposed to recruit PPIL1 to the spliceosome. May be involvedin cyclin-D1/CCND1 mRNA stability through the SNARP complex whichassociates with both the 3'end of the CCND1 gene and its mRNA.Involved in transcriptional regulation. Modulates TGF-beta-mediated transcription via association with SMAD proteins, MYOD1-mediated transcription via association with PABPN1, RB1-mediatedtranscriptional repression, and retinoid-X receptor (RXR)- andvitamin D receptor (VDR)-dependent gene transcription in a cellline-specific manner probably involving coactivators NCOA1 andGRIP1.
