Cleaves the gamma-glutamyl bond of extracellularglutathione (gamma-Glu-Cys-Gly), glutathione conjugates, and othergamma-glutamyl compounds. The metabolism of glutathione releasesfree glutamate and the dipeptide cysteinyl-glycine, which ishydrolyzed to cysteine and glycine by dipeptidases. In thepresence of high concentrations of dipeptides and some aminoacids, can also catalyze a transpeptidation reaction, transferringthe gamma-glutamyl moiety to an acceptor amino acid to form a newgamma-glutamyl compound. Initiates extracellular glutathione (GSH)breakdown, provides cells with a local cysteine supply andcontributes to maintain intracellular GSH level. It is part of thecell antioxidant defense mechanism.
