KPNA4

O00629

Receptor tyrosine kinase which binds promiscuously GPI-anchored ephrin-A family ligands residing on adjacent cells, leading to contact-dependent bidirectional signaling intoneighboring cells. The signaling pathway downstream of thereceptor is referred to as forward signaling while the signalingpathway downstream of the ephrin ligand is referred to as reversesignaling. Among GPI-anchored ephrin-A ligands, EFNA5 mostprobably constitutes the cognate/functional ligand for EPHA5.Functions as an axon guidance molecule during development and maybe involved in the development of the retinotectal, entorhino-hippocampal and hippocamposeptal pathways. Together with EFNA5plays also a role in synaptic plasticity in adult brain throughregulation of synaptogenesis. In addition to its function in thenervous system, the interaction of EPHA5 with EFNA5 mediatescommunication between pancreatic islet cells to regulate glucose-stimulated insulin secretion (By similarity). {ECO:0000250}. Functions in nuclear protein import as an adapterprotein for nuclear receptor KPNB1. Binds specifically anddirectly to substrates containing either a simple or bipartite NLSmotif. Docking of the importin/substrate complex to the nuclearpore complex (NPC) is mediated by KPNB1 through binding tonucleoporin FxFG repeats and the complex is subsequentlytranslocated through the pore by an energy requiring, Ran-dependent mechanism. At the nucleoplasmic side of the NPC, Ranbinds to importin-beta and the three components separate andimportin-alpha and -beta are re-exported from the nucleus to thecytoplasm where GTP hydrolysis releases Ran from importin. Thedirectionality of nuclear import is thought to be conferred by anasymmetric distribution of the GTP- and GDP-bound forms of Ranbetween the cytoplasm and nucleus. In vitro, mediates the nuclearimport of human cytomegalovirus UL84 by recognizing a non-classical NLS. In vitro, mediates the nuclear import of humancytomegalovirus UL84 by recognizing a non-classical NLS.
