Histone deubiquitinating component of the transcriptionregulatory histone acetylation (HAT) complex SAGA. Catalyzes thedeubiquitination of both histones H2A and H2B, thereby acting as acoactivator. Recruited to specific gene promoters by activatorssuch as MYC, where it is required for transcription. Required fornuclear receptor-mediated transactivation and cell cycleprogression. {ECO:0000269|PubMed:18206972,ECO:0000269|PubMed:18206973, ECO:0000269|PubMed:18469533}. Component of the origin recognition complex (ORC) thatbinds origins of replication. DNA-binding is ATP-dependent. Thespecific DNA sequences that define origins of replication have notbeen identified yet. ORC is required to assemble the pre-replication complex necessary to initiate DNA replication. Doesnot bind histone H3 and H4 trimethylation marks H3K9me3, H3K27me3and H4K20me3. {ECO:0000269|PubMed:22427655}.
