SARNP

P82979

Acts as E3 ubiquitin-protein ligase and regulates thedegradation of target proteins. Polyubiquitinates MYD88.Negatively regulates MYD88-dependent production of proinflammatorycytokines. Can promote TRIF-dependent production of type Iinterferon and inhibits infection with vesicular stomatitis virus(By similarity). Promotes also activation of TBK1 and IRF3.Involved in the ubiquitination of erythropoietin (EPO) andinterleukin-3 (IL-3) receptors. Thus, through maintaining basallevels of cytokine receptors, RNF41 is involved in the control ofhematopoietic progenitor cell differentiation into myeloerythroidlineages (By similarity). Contributes to the maintenance ofsteady-state ERBB3 levels by mediating its growth factor-independent degradation. Involved in the degradation of theinhibitor of apoptosis BIRC6 and thus is an important regulator ofcell death by promoting apoptosis. Acts also as a PRKN modifierthat accelerates its degradation, resulting in a reduction of PRKNactivity, influencing the balance of intracellular redox state. The RNF41-PRKN pathway regulates autophagosome-lysosome fusionduring late mitophagy. Mitophagy is a selective form of autophagynecessary for mitochondrial quality control (PubMed:24949970).{ECO:0000250, ECO:0000250|UniProtKB:Q8BH75,ECO:0000269|PubMed:12411582, ECO:0000269|PubMed:14765125,ECO:0000269|PubMed:15632191, ECO:0000269|PubMed:17210635,ECO:0000269|PubMed:18541373, ECO:0000269|PubMed:19483718,ECO:0000269|PubMed:24949970}. Binds both single-stranded and double-stranded DNA withhigher affinity for the single-stranded form. Specifically bindsto scaffold/matrix attachment region DNA. Also binds single-stranded RNA. Enhances RNA unwinding activity of DDX39A. Mayparticipate in important transcriptional or translational controlof cell growth, metabolism and carcinogenesis. Component of theTREX complex which is thought to couple mRNA transcription, processing and nuclear export, and specifically associates withspliced mRNA and not with unspliced pre-mRNA. TREX is recruited tospliced mRNAs by a transcription-independent mechanism, binds tomRNA upstream of the exon-junction complex (EJC) and is recruitedin a splicing- and cap-dependent manner to a region near the 5'end of the mRNA where it functions in mRNA export to the cytoplasmvia the TAP/NFX1 pathway. The TREX complex is essential for theexport of Kaposi's sarcoma-associated herpesvirus (KSHV)intronless mRNAs and infectious virus production.{ECO:0000269|PubMed:15338056, ECO:0000269|PubMed:17196963,ECO:0000269|PubMed:20844015}.
