Metallocarboxypeptidase that mediates deglutamylation oftarget proteins. Catalyzes the deglutamylation of polyglutamateside chains generated by post-translational polyglutamylation inproteins such as tubulins. Also removes gene-encodedpolyglutamates from the carboxy-terminus of target proteins suchas MYLK. Acts as a long-chain deglutamylase and specificallyshortens long polyglutamate chains, while it is not able to removethe branching point glutamate, a process catalyzed by AGBL5/CCP5.{ECO:0000250|UniProtKB:Q09M05}.
