As a component of the GATOR1 complex functions as aninhibitor of the amino acid-sensing branch of the TORC1 pathway.The GATOR1 complex strongly increases GTP hydrolysis by RRAGA andRRAGB within RRAGC-containing heterodimers, thereby deactivatingRRAGs, releasing mTORC1 from lysosomal surface and inhibitingmTORC1 signaling. The GATOR1 complex is negatively regulated byGATOR2 the other GATOR subcomplex in this amino acid-sensingbranch of the TORC1 pathway. {ECO:0000269|PubMed:23723238}. Ubiquitin-conjugating enzyme E2 that specifically actswith HECT-type and RBR family E3 ubiquitin-protein ligases. Doesnot function with most RING-containing E3 ubiquitin-proteinligases because it lacks intrinsic E3-independent reactivity withlysine: in contrast, it has activity with the RBR family E3enzymes, such as PRKN and ARIH1, that function like function likeRING-HECT hybrids. Accepts ubiquitin from the E1 complex andcatalyzes its covalent attachment to other proteins. In vitrocatalyzes 'Lys-11'-linked polyubiquitination. Involved in theselective degradation of short-lived and abnormal proteins. Down-regulated during the S-phase it is involved in progression throughthe cell cycle.
