Major role in the synthesis of nucleoside triphosphatesother than ATP. The ATP gamma phosphate is transferred to the NDPbeta phosphate via a ping-pong mechanism, using a phosphorylatedactive-site intermediate. May be a allosteric regulator of serine/threonine-protein phosphatase 2A (PP2A). Isoform 1 inhibits catalyticactivity of the PP2A(D) core complex in vitro. The PP2A(C):TIPRLcomplex does not show phosphatase activity. Acts as negativeregulator of serine/threonine-protein phosphatase 4 probably byinhibiting the formation of the active PPP4C:PPP4R2 complex; thefunction is proposed to implicate it in DNA damage response bypromoting H2AFX phosphorylated on Ser-140 (gamma-H2AFX).
