KPNA6 | TRAIP |
Functions in nuclear protein import as an adapterprotein for nuclear receptor KPNB1. Binds specifically anddirectly to substrates containing either a simple or bipartite NLSmotif. Docking of the importin/substrate complex to the nuclearpore complex (NPC) is mediated by KPNB1 through binding tonucleoporin FxFG repeats and the complex is subsequentlytranslocated through the pore by an energy requiring, Ran-dependent mechanism. At the nucleoplasmic side of the NPC, Ranbinds to importin-beta and the three components separate andimportin-alpha and -beta are re-exported from the nucleus to thecytoplasm where GTP hydrolysis releases Ran from importin. Thedirectionality of nuclear import is thought to be conferred by anasymmetric distribution of the GTP- and GDP-bound forms of Ranbetween the cytoplasm and nucleus. {ECO:0000269|PubMed:10523667}. | E3 ubiquitin ligase acting as a negative regulator ofinnate immune signaling. Inhibits activation of NF-kappa-Bmediated by TNF. Negatively regulates TLR3/4- and RIG-I-mediatedIRF3 activation and subsequent IFNB1 production and cellularantiviral response by promoting 'Lys-48'-linked polyubiquitinationof TNK1 leading to its proteasomal degradation (By similarity)(PubMed:22945920). Involved in response to genotoxic lesionsduring genome replication. Promotes H2AX and RPA2 phosphorylationafter replication-associated DNA damage and assists forkprogression at UV-induced replication-blocking lesions during Sphase (PubMed:26595769). Has also been proposed to play a role inpromoting translesion synthesis by mediating the assembly of 'Lys-63'-linked poly-ubiquitin chains on the Y-family polymerase POLNin order to facilitate bypass of DNA lesions and preserve genomicintegrity (PubMed:24553286). The function in translesion synthesisis controversial (PubMed:26595769). {ECO:0000250|UniProtKB:Q8VIG6,ECO:0000269|PubMed:22945920, ECO:0000269|PubMed:24553286,ECO:0000269|PubMed:26595769, ECO:0000303|PubMed:26595769}. |
