Receptor for Wnt proteins. Most of frizzled receptorsare coupled to the beta-catenin (CTNNB1) canonical signalingpathway, which leads to the activation of disheveled proteins,inhibition of GSK-3 kinase, nuclear accumulation of beta-catenin(CTNNB1) and activation of Wnt target genes. Plays a critical rolein retinal vascularization by acting as a receptor for Wntproteins and norrin (NDP). In retina, it can be both activated byWnt protein-binding, but also by a Wnt-independent signaling viabinding of norrin (NDP), promoting in both cases beta-catenin(CTNNB1) accumulation and stimulation of LEF/TCF-mediatedtranscriptional programs. A second signaling pathway involving PKCand calcium fluxes has been seen for some family members, but itis not yet clear if it represents a distinct pathway or if it canbe integrated in the canonical pathway, as PKC seems to berequired for Wnt-mediated inactivation of GSK-3 kinase. Bothpathways seem to involve interactions with G-proteins. May beinvolved in transduction and intercellular transmission ofpolarity information during tissue morphogenesis and/or indifferentiated tissues. As part of the Ragulator complex it is involved in aminoacid sensing and activation of mTORC1, a signaling complexpromoting cell growth in response to growth factors, energylevels, and amino acids. Activated by amino acids through amechanism involving the lysosomal V-ATPase, the Ragulatorfunctions as a guanine nucleotide exchange factor activating thesmall GTPases Rag. Activated Ragulator and Rag GTPases function asa scaffold recruiting mTORC1 to lysosomes where it is in turnactivated. LAMTOR1 is directly responsible for anchoring theRagulator complex to membranes. Also required for lateendosomes/lysosomes biogenesis it may regulate both the recyclingof receptors through endosomes and the MAPK signaling pathwaythrough recruitment of some of its components to late endosomes.May be involved in cholesterol homeostasis regulating LDL uptakeand cholesterol release from late endosomes/lysosomes.
